Lain Iwakura The titular character of the series. Lain is a fourteen-year-old girl who uncovers her true nature through the series. She is first depicted as a shy junior high school student with few friends or interests. She later grows multiple bolder personalities, both in the physical world and the Wired, and starts making more friends. As the series progresses, she eventually comes to discover that she is, in reality, merely an autonomous, sentient computer program in the physical and corporeal form of a human being, designed to sever the invisible barrier between the Wired and the real world. In the end, Lain is challenged to accept herself as a de facto goddess for the Wired, having become an omnipotent and omnipresent virtual being with worshipers of her own, as well as an ability to exist beyond the borders of devices, time, or space.

Ruri Gokou Ruri Gokou—perhaps better known as her alter-ego Kuroneko (“Black Cat”)—is a member of the All Together, Otaku Girls online community that Saori manages. She goes to an offline meet-up where she meets both Saori and Kirino in person. Like Kirino, she remains aloof during this meeting, and it isn't until after the meeting at Saori's post-gathering that she is formally introduced. Kuroneko is a fan of teenage fantasy anime and is sickened by cute anime, like Meruru. Naturally, this precipitates several arguments between Kirino and her, but they still manage to maintain a friendship despite their differences. Kuroneko is shown to care greatly for her friends despite her cold attitude towards people of “the mortal world.”

Kamina Kamina (カミナ) is a young man from Giha, a subterranean village, who dreams of leaving the underground and going to the surface, a place which he alone believes he saw as a child. Possessing a seemingly arrogant personality, Kamina is very passionate about his goals and will pursue them without a trace of fear, hesitation, or sometimes sense. This attitude often manifests itself in dramatic speeches which have the power to rally all those who hear them to his call. As his first act of notoriety, Kamina forms "Team Gurren" and convinces his "blood-brother," Simon, to help him with his plan to drill through the roof of the village and get to the world above. However, the plan fails. Kamina willingly takes the blame for disrupting the peace and is jailed by the village chief. At night, Simon breaks him out to show him an extraordinary artifact he unearthed, only to be interrupted by a large Gunmen that crashes through the village's ceiling. A young girl sharpshooter named "Yoko" descends from the surface and ends up joining Kamina and Simon as they pilot the newly unearthed Gunmen Kamina names "Lagann". Together, they defeat the invading Gunmen and reach the surface in the process.

Takashi Natsume Natsume Takashi is the main protagonist of the series who, like his deceased grandmother Natsume Reiko, has the ability to see ayakashi (spirits). He inherited the Book of Friends from her, a book of contracts binding ayakashi to servitude when they were defeated by Reiko. Due to Natsume's strong resemblance to his grandmother, he is often mistaken for her and is pursued by ayakashi who hate Reiko but yearn for the Book of Friends. To rid himself of these troubles, Natsume resolved to end the book by returning all the contracted names to their former owners. He is accompanied by the youkai Madara, nicknamed "Nyanko-sensei" (or simply "sensei"), promising him possession of the book once he dies. In return, the creature must serve as his bodyguard until then. On the other hand, most of the ayakashi let Natsume see their memories, so that he can understand them better. Natsume can also see their memories when he releases a yokai's name. When Natsume is completely frustrated over a problem and falls asleep in class, he actually sleep-draws in his notebook about whatever is bothering him at the time.
